First question: What device do you have?
Second question: By "with unlock" do you just mean Chevron unlock, official Dev unlock, or Interop unlock? The instructions for the first and third are device-specific, but flashing an official ROM will always remove them. For the second, just unlock your phone again using the WP7 device registration tool.
As for unlocking a locked phone that is already on Mango (and that you don't want to roll back), there are a few options for dev unlock:
Buy a marketplace dev account from MS ($100).
Get a free marketplace dev account through DreamSpark (intended for students).
Put a CustClear.provxml file in the \Windows\ folder (must be done prior to upgrading), upgrade to Mango *WITHOUT* the latest HTC update (if you flash a ROM instead of upgrading in place, that file will be removed), and run Connection Setup.
Wait "a few weeks" until ChevronWP7 Labs is available, and pay the $9 for them to unlock your phone.
Note that if your phone is an HTC phone, as it seems, you need to be very wary of the latest HTC update or you won't be able to Interop-unlock at all, and the unofficial ways to Dev-unlock will mostly stop working. Don't install that update until you're fully unlocked. Even then, you'll lose access to the filesystem and registry DLLs.

if you are not unlocked and upgrade to Mango you will have a hard time unlocking it within Mango (and won't be able to sideload). Unlock in pre-NoDo and then carry the unlock over to NoDo and Mango following the guide. yes, people have successfully carried their Chevron unlock over to NoDo and then Mango. You need to make sure that it cannot relock when you connect to Zune. The tools listed allow you to make the reg changes to do that.

What should I do for preventing my device being relocked during the ongoing official Mango update?
Is there already some solution, or will it be after the official Mango release will appear in Zune?

I think we are talking about two different kinds of unlocking. I am referring to unlocking the phone for sideloading homebrew apps. If you upgrade to Mango you might not be able to sideload homebew apps that allow you to customize your phone until the gurus come up with a solution for Mango. Then there is SIM unlock to be able to use a device with a different SIM. I think that is what you are referring to. That will not change for you if you upgrade to Mango.

If you follow Ansar's tutorial you will see that there are preventions you can take to stay unlocked. You basically finished part A of his tutorial and can continue to part B with prerequisites for Mango upgrade.
B. MangoB2 Update Guide: How To Pass ChevronWP7 from OS 7392 [NoDo] to OS 7403 and 7712
In your case 7712 will be 7720.68 once it is released.
When you have all prerequisits finished from Ansar's post you stop there and are ready for official Mango once it comes out.
Reports from people who were NoDo unlocked and proceeded to Mango are mixed. Some kept the unlock some didn't. If it doesn't stay unlocked you have to have the files ready on the device to be able to unlock it without going back to NoDo. This is what Ansar's post explains. For the HTC you will put a CustClear.provxml file in place that you can deploy to unlock it.
This post also explains how to keep it unlocked. You follow the guide to the point at which it does the actual Mango upgrade. That is where you stop and wait for the official one.
But even if your device is locked in Mango you can go back to a NoDo restore point you created before you went to Mango. All explained in the guides/tutorials above.

those who just upgraded. I myself just upgraded but luckily for LG users there is an LG phone tool on the device that allows to modify the registry at any stage.

Today I have succesfuly registered my ISIC student card with validity until december this year into the Microsoft Developer Account.
Now I have Chevron unlocked NoDo WP7 (HTC Trophy) with some modifications to prevent it from relocking during sync etc. I would like to unlock it via official Developer Account I now have.
1) If am not wrong - I should HR my phone, it will go to NoDo locked and then I can use application "Windows Phone Developer Registration" which will unlock my device and it will stay unlocked also after Mango update?
2) Is it possible by developer unlocked device to modify registry entries? I need to add one entry, so I will have Mango in my native language after update.
3) After one year, will it automatically lock my device and I will need to perform new ISIC validation, which will be in that day already expired?
4) If it will expire, will it hard reset or block my phone or will it stay in way it is, but it will not only be possible to side load xap's?

I need to unlock my HTC Radar which is with WP 7.5 Mango OS version: 7.10.7740.16. I read a lot of posts and can not find solution. I wait till ChevronWP7 labs provide unlock tokens again. Yesterday I buy token follow the procedure and I receive message that my phone is unlocked. After that I try to use Tom XAP installer to install some Apps on my phone but it didn't work it says that my device is not unlocked. After that I try do deploy an Apps buy SDK tools: Application Deployment but again I receive an error that is listed here in the forums. I have to check if my device is unlocked or not but I am not quite sure how to do that. I start Windows Phone Developer Registration and it says that I am already registered and give me option to Unregistered is that mean I am with unlocked phone ? I want do deploy some apps to my phone please help. I read some treads here that says for my phone still not possible to unlock the phone because use driversv2 is this true or I just can not find the way to unlock my phone ? Also whit this ChevronWP7 unlocker how much apps I can deploy ?
You might have to interop unlock your phone. I'm just guessing her but it's worth a shot.

Please suggest me what can I do to install more apps. I need to install Bulgarian keyboard I try but it can not install the XAP file also I want to install wi fi and Bluetooth fast switches, I need navigation program too and I see Navigon that may be will work fine.
Interop unlock will only be needed for some homebrew apps. You should be able to install other apps without interop unlock.
I think second Gen phones like Radar cannot be interop unlocked as of now.

I dont know what interop-unlock means or what are the difference with dev unlocked. My device is locked. I would like to install applications like those I see here on the forums. One year ago, when I unlocked my device, pre-mango, I used Tom Xap Installer to do so.
Okay, then you need dev-unlock. There is only two ways for stock ROMs on the HTC phones and one of those is temporarily unavailable (that one being buying ChevronWP tokens that allow you to unlock; they are out of tokens from MS so unless you bought one you have to wait).
The other one is sign-up with MS App Hub and pay $99/yr unless you are a student (then you can get it for free but takes a little proof you are a student). This is the only way for a Mango Stock ROM.
Now, if your HTC Trophy is NOT Verizon, there are custom ROMs and methods to flash your phone with an unlocked custom ROM, but this wipes your phone and takes a little know-how.
This is a quick summary and there are many details on all of this in various threads (and many of them). But in summary, if you want to keep your Stock ROM/Phone, the only way right now is sign up for App Hub at MS or wait for ChevronWP Labs to get more tokens.

Is still one more way - downgrade your SPL and install RSPL/HSPL and install the fully unlocked costom ROM for your device.

3. Custom ROMs for Trophy: http://forum.xda-developers.com/forumdisplay.php?f=812 select one and install, when you get bored you can install other ROM from that forum chapter.
I have some question about developer unlocking.
Since I living in the country that Windows Phone 7 didn't had its local keyboard (yet), I have to install app like native keyboard for WP7 which requires developer unlocked phone and I won't pay $100 per year just to install only 1 app. Also, I know how to unlock because I used to unlock my friend's phone.
1. Is the app which installed via sideload still work after the phone has been re-locked?
2. Is the same app from question #1 still work after I installed latest update? For example, I installed some app via sideload into the phone which has Windows Phone 7 pre-NoDo update, then update that phone to latest update.
1) No, sideloaded apps will stop working if the phone is re-locked. You'll get an (inaccurate but scary) message saying the app has been "revoked by Microsoft".
2) Installing the Mango update will re-lock your phone. For people with legit unlocks, this is trivial to fix. For everybody else... it depends on what phone you're on. Dell can't be unlocked by hacks at all, right now. LG can be unlocked easily via the built-in registry editor. Samsung can be unlocked easily via WindowBreak. HTC can't currently use WindowBreak but that should change soon, although it will be more complicated than for Samsung.
Also, the original ChevronWP7 Unlocker doesn't work anymore, even on pre-NoDo ROMs; the certificate has expired. You'd need to set the clocks way back.

For HTC, the only current option is to get an AppHub account (free to students, $99/year otherwise). Also, your bootloader version indicates that you've installed an update that will prevent your phone from being "interop-unlocked" so high-privilege XAPs won't be available until you downgrade your phone's firmware.
...you could find some help in this thread , but since you say that you don't understand too much you may better leave it. Otherwise you might break your phone.
You might also wait for Microsoft to enable Chevron unlock tokens again - which I don't think is very realistic: https://labs.chevronwp7.com
Unlocking your phone via chevron is very simple and doesn't need any know-how, but it won't (just like with the developer account) allow you to use every .XAP application.
