On a stock g2x rom and kernal how should I partition my SD card? Ntfs, fat, ext2, ext3, ext4, Linux swap, or unformatted?

Depends on the ROM you're using. With stock and CM7 you're fine with fat32.

The main benefits that ext4 has over ext3 are:
faster timestamping faster file system checking journaling checksums extents (basically automatic space allocation to avoid fragmentation)

There is a plethora of information on this. Please search first.
Ext3 and ext4 are linux filesystems that are unrelated to Android in general. You can probably find out more information on the pros/cons of each by searching specifically as a linux search rather than an android specific search.

I too would like an answer to this question. I am running rooted stock 2.3.4.......
Are there any that read them other than stock?
I'm talking Jelly Bean. I'm on Clean Rom 6 and looking for options to over clock.
Ziggys supports 64 formatted as fat32 I think. Comes bundled with synergy. Haven't tried it yet though, only have a 32 gb card.

I'm trying to create another fat32 partition for apps using mini tool partitio in my friend's htc pico but link2sd always chooses first partition(the one for music,videos). I did unlock bootloader and root. Please help!

Well.... make it ext, maybe the kernel is confusing things, also you can try with mounts2sd.
My friends phone is stock rom so ext is not supportes

Garuda or cramium for stock rom
And use m2sd and make sd-ext partition as ext2 or ext3
