If you have new hboot (2.00.002) and you are S-OFF, it means your phone has hardware S-OFF, using XTC clip. You don't have to unlock it. You can do right about everything with your phone without unlocking it.
So why is this

Try 'fastboot oem unlock'
Some first gen phone's were s-off to start with. Don't quote me on that, I'm guessing.

Because s-off/s-on and locked/unlocked are 2 different things. Trust me being s-off'ed by xtc clip is more then enough, you can install anything you want. I have a phone like that. Don't unlock, there is no need for that. You can flash any RUU if you want so and still stay s-off and then flash any custom rom, and then RUU again... I've done that all more then once.

Since your phone already is s-off,
You can root it now using a tool like superoneclick or something else.

Agree with the above.
What I personaly did is installed latest RUU 2.10.401.9 just to have the latest hboot and radio (since I have xtc clip s-off, I stayed s-off). Then rooted it with one click tool, flashed latest 4ext recovery and installed my favorite custom rom.

Is it necessary to take nandroid backup before rooting or we can simply backup our appz

Are you kidding?? Already told you.
If it's already s-off, root it with superoneclick, install recovery like 4ext or cwm, backup everything and flash roms like it's going crazy.
Search the forums, it's written a hundret times everywhere!#!

Dude, you know, I'm not gonna do the simple search work for you. I tried to help you but I have other things to do aswell. If you're not able to find the mentioned stuff by yourself, you should probably not play around with your device...

Sigh
If you are s-on use the juopuntbear method to s-off or if you did something else to brick it AS WELL look for a guide by dexter.

i want to root my sensation xe and try some custom roms, i've rooted my Hero once and thought rooting my sensation would not be so difficult.
Here is my problem: i unlocked using HTCDev but s-on is still there, HBOOT version = 1.29.0000 and firmware is 3.33.401.106
is there any noob friendly way i can root my sensation xe at this time? i tried finding one but they all refer to firmware 3.32.xxx.xxx.
idont know what to do, please help if you can :fingers-crossed:

All the ones that refer to 3.32.xxx.xx also works for 3.33.xxx.xx, so go ahead and use them

I think he means that you should downgrade the hboot but it is not possible with an s-on device.
To s-off your device you can use this tutorial http://forum.xda-developers.com/showthread.php?t=1614617
Root your device with this tool http://forum.xda-developers.com/showthread.php?t=1668276
But be careful and read the readme, no one takes responsibility if you brick your device.

I got i rooted, but juonuputbear corrupted my sdcard, it fixed now and I'm running CM10

Yes there is a easy way of doing it you could flash a ruu that has the boot 1.17 and then head on over to alpha revolutionary and then s off and then root and then head over to 4ext.net download your new custom recovery and set it up and then upgrade your firmware to 3.32 or 3.33 you will then have hboot 1.27 next flash a Rom
